Studies on the shigella flexneri phage


Abstract
The author has studied on the activity of shigella flexneri phage which is suspended in various fluids and kept long time at various temperatures and obtained the following results.
When the phage is suspended in the neutral distilled water, 0.85 % saline, 1. 0 -peptone water, common broth, 0.5 % glucose broth and 0. 01 % gelatin broth respectively and kept at 4¡ÆC, the time necessary for about 50 % inactivation of adding phage was about 4 weeks, 2 weeks, 7 months, 7 months, 8 weeks and 4 months respectively, and when keep at room temperature, the time was about 3 weeks, 4 days, 4 weeks, 3 weeks, one week and 2 weeks respectively, and when keep at 37¡ÆC, it was about 5 days, 2 days, 4 days, 4 days, one day and 2 days respectively. And then, when keep at 50¡ÆC, about 80 % of the phage suspended in broth was inactivated for one day and the phage is seemed to be a little stable in the presence of small dosis of Mg++.
The phage was most stable when the pH of phage suspension is neutral and the effect of pH on phage activity was different respectively according to the kinds of fluid and temperature for keeping.
The period for discovering of phage was longer in the higher concentration of phage than lower concentration.
The increasing grade of phage inactivating rate according to increasing of temperature was more remarkable when the phage is suspended in complicated fluid than simple one.
